Regioselective Acylation of Nucleosides Catalyzed by Candida Antarctica Lipase B: Enzyme Substrate Recognition

The substrate recognition of Candida antarctica lipase B (CAL-B) in the acylation of nucleosides was revealed through rational substrate engineering for the first time. CAL-B displayed lower activities and excellent 5[prime]-regioselectivities (94 to >99 %) in the acylation of ribonucleosides 1f-1j as compared to those in the acylation of 2[prime]-deoxynucleosides 1a-1e. The excellent regioselectivities might be attributed to the favorable steric hindrance of the 2[prime]-hydroxy group present in ribonucleosides 1f-1j, which destabilizes the conformation of the 3[prime]-acylation transition state in the acylation of 1f-1j and thus reduces the amount of the minor regioisomer. The study of the acyl chain-length specificity showed that CAL-B was more specific toward short-chain fatty acid vinyl esters, especially vinyl butyrate. (© Wiley-VCH Verlag GmbH & Co. Preparation of the First Bench-Stable Phenyl Selenolate: an Interesting "On Water" Nucleophilic Reagent

In this communication we report the synthesis and the characterization of the first solid and air-stable selenolates, starting from commercially available phenylselenenyl halides and elemental zinc. These reagents were efficiently employed in the ring opening of epoxides as well as in other nucleophilic substitution and addition reactions showing an unexpected rate acceleration in water suspension at room temperature. Palladium-Catalyzed Amination and Sulfonylation of 5-Bromo-3-[2-(diethylamino)ethoxy]indoles to Potential 5-HT6 Receptor Ligands

A general and efficient palladium-catalyzed amination of 5-bromo-3-[2-(diethylamino)ethoxy]indoles has been developed. Best results are obtained in the presence of Pd(OAc)2 and 2-[di(1-adamantyl)phosphanyl]-1-phenylpyrrole as ligand. Subsequent sulfonylation gave novel indole derivatives, which are of interest as potentially biological active 5-HT6 receptor ligands.(© Wiley-VCH Verlag GmbH & Co. Straightforward Strategy for the Stereoselective Synthesis of Spiro-Fused (C-5)Isoxazolino- or (C-3)Pyrazolino-(C-3)quinolin-2-ones from Baylis-Hillman Adducts by 1,3-Dipolar Cycloaddition and Reductive Cyclization

A straightforward and general approach for the stereoselective synthesis of spiro-fused (C-5)isoxazolino- or (C-3)pyrazolino-(C-3)quinolin-2-ones from the adducts offorded from the Baylis-Hillman reaction of 2-nitrobenzaldehyde and ethyl acrylate by sequential 1,3-dipolar cycloaddition and reductive cyclization is presented. It was found that the reductive cyclization of the isoxazoline derivatives proceeded efficiently in the presence of In/HCl, whereas similar reductions of pyrazolines gave better yields when carried out in the presence of an Fe/AcOH mixture. However, similar attempts employing the Baylis-Hillman adduct of 2-nitrobenzaldehyde and methyl vinyl ketone did not yield the desired compounds.(© Wiley-VCH Verlag GmbH & Co. Microwave-Assisted Stereoselective One-Pot Synthesis of Symmetrical and Unsymmetrical 2,5-Diketopiperazines from Unprotected Amino Acids

The facile condensation of unprotected amino acids by a phosphite-promoted one-step coupling reaction is a highly efficient synthesis to generate stereoselective and optically pure symmetrical and unsymmetrical functionalized diketopiperazines. The use of microwaves enhanced by small amounts of ionic liquid is accompanied by significant improvement in reaction times and yields. Taxpropellane: A Novel Taxane with an Unprecedented Polycyclic Skeleton from the Needles of Taxus canadensis

Taxpropellane, the first example of a taxane with an unprecedented 6/5/5/6/4/5-membered ring hexacyclic skeleton ([3.3.2]propellane), was isolated from the needles of Taxus canadensis. Oxime-Based Synthesis of New Chromogenic and Fluorogenic Oligosaccharides

A new simple approach for the synthesis of labelled oligosaccharides is described. Free synthetic carbohydrate recognition building blocks were conjugated with fluorogenic and chromogenic dyes under mild experimental conditions by using oxime ligation. The resulting oligosaccharide probes presenting well-defined anomer configuration might find broad interests for recognition studies with a large panel of carbohydrate-binding proteins. Synthesis and Electrochemical Properties of Structurally Modified Flavin Compounds

Four structurally modified flavin compounds have been synthesized and characterized for their redox potential by chemical reduction with sodium dithionite. Besides the previously reported 1- and 5-deazariboflavin, a 7,8-didemethyl derivative and an 8-isopropylriboflavin have been obtained. The synthesis of these compounds started in all cases from appropriately substituted anilines that were condensed with the ribityl chain, followed by completion of the annealed three-ring structure. The didemethyl- and the isopropyl compounds gave absorption maxima similar to riboflavin (436 and 448 nm, respectively), whereas 1-deazariboflavin showed a bathochromically shifted absorption ([lambda]max = 537 nm), and that of 5-deazariboflavin was hypsochromically shifted ([lambda]max = 400 nm). The midpoint potentials (E0[prime]) of the four modified flavin compounds were determined by potentiometric titration, using riboflavin as a reference compound. Both alkyl-modified flavins showed slightly less negative midpoint potentials, whereas both deaza compounds had more negative midpoint values compared to the reference compound. Synthesis and Glycosidase Inhibitory Study of New Polyhydroxylated Indolizidines

The synthesis of two polyhydroxylated indolizidines as potenticial glycosidase inhibitors is reported. The piperidine ring was formed by an intramolecular Mannich-type reaction between ethyl trans-4-oxo-2-butenoate and the two [beta]-amino ketones (-)-1-(2-methyl-1,3-dioxan-2-yl)propan-2-amine and(+)-1-(benzyloxymethyl)-2-(2-methyl-1,3-dioxan-2-yl)ethylamine. The synthesis of this amine was performed in eight steps from L-aspartic acid. The key steps of the formation of the framework involved dihydroxylation, nucleophilic substitution, and reduction. The last hydroxy group was introduced by hydrolysis of the acetal moiety followed by reduction of the resulting ketone. Lewis Acid Catalyzed Reactions of Donor-Acceptor Cyclopropanes with Anthracenes

The reactions of 2-aryl-1,1-cyclopropane diesters with anthracene derivatives were studied for the first time. Three kinds of reaction products are formed depending on the nature of the aryl group and the substituents in the anthracene. The first one is the product of [4+3] cycloaddition of cyclopropane to a diene moiety of anthracene. The second one also has a seven-membered ring and is formed by the Lewis acid catalyzed electrophilic attack of cyclopropane onto the C9 atom of anthracene followed by intramolecular attack of the formed arenonium ion on aryl group of the starting cyclopropane. Directed (R)- or (S)-Selective Dynamic Kinetic Enzymatic Hydrolysis of 1,2,3,4-Tetrahydroisoquinoline-1-carboxylic Esters

The first synthesis of both enantiomers of 6,7-dimethoxy-1,2,3,4-tetrahydroisoquinoline-1-carboxylic acid was accomplished through dynamic kinetic resolution in procedures based on CAL-B- or subtilisin Carlsberg-catalysed enantioselective hydrolysis of the corresponding ethyl esters in aqueous NH4OAc buffer at pH 8.5. The products were obtained with high enantiopurity (92-93 % ee) in good yields (85-92 %). (R)-1,2,3,4-Tetrahydroisoquinoline-1-carboxylic acid was obtained with high enantiopurity (98 % ee) and in good yield (85 %) in a CAL-B-catalysed process, under similar conditions.(© Wiley-VCH Verlag GmbH & Co. Synthesis of thienyl analogues of PCBM and investigation of morphology of mixtures in P3HT

Abstract

Novel [6,6]-phenyl-C61-butyric acid methyl ester (PCBM) analogues containing benzo[b]thiophene (3a, 3b) and thieno[3,2-b]thiophene (3c, 3d) were synthesized and characterized. The morphology of the thin films prepared from the mixtures of these methanofullerenes with regioregular poly(3-hexylthiophene) (P3HT) was investigated by AFM measurement and UV-Vis absorption spectroscopy. A solubility test of these methanofullerenes was performed by using dichloromethane as a solvent. es-TThCBM (3d) exhibited 1.4 times greater solubility in dichloromethane than PCBM.
